Background High-temperature requirement A serine peptidase 1 (HTRA1, PRSS11) is a trypsin-like serine protease involved in neuronal maturation, protein degradation, apoptosis, and the response to cellular stress. HTRA family members contain N-terminal IGFBP and Kazal-like domains, a serine protease domain, and a C-terminal PDZ domain (1). HTRA1 can be secreted and autocatalyzed (2) or retained in the cell and processed to shorter cytoplasmic- and nuclear-localized forms (3). Ser328Ala mutants lack both proteolytic and autocatalytic activity (2). HTRA1 regulates the extracellular matrix (ECM) by degrading aggrecan, collagen, decorin, fibromodulin, and fibronectin (4,5), while also breaking down signaling molecules such as TGF-β family members (6,7) and FGF8 (8). In the brain, HTRA1 degrades β-Amyloid and aggregated fibrillar tau (9,10). Elevated HTRA1 can contribute to osteoarthritis (5,11) and age-related macular degeneration (12). HTRA1 loss induces EMT and breast cancer cell invasion (13).
