Background Hypoxanthine phosphoribosyltransferase 1 (HPRT1) is a key enzyme in the purine salvage pathway. Studies show that, in the presence of the PI3K/mTOR dual inhibitor gedatolisib, reduced HPRT1 expression leads to decreased survival rate of small-cell lung carcinoma (SCLC) cells, suggesting that HPRT1 plays a role in SCLC cell sensitivity to gedatolisib (1). Furthermore, electron transport chain (ETC) inhibition suppresses purine synthesis and enhances the purine salvage pathway. HPRT1 was shown to be important for non-small cell lung cancer (NSCLC) growth when ETC is inhibited (2). In addition, HPRT1 is involved in the metabolism of temozolomide (TMZ) to activate AMPK and promote chemoresistance of glioblastomas (GBMs) (3).
