Background Mammalian alkaline phosphatases (ALPs) are highly conserved zinc-containing allosteric enzymes that hydrolyze and transphosphorylate a wide range of compounds (1). There are four known human alkaline phosphatase isozymes: ALPL (tissue-nonspecific; bone/liver/kidney), ALPP (placental), ALPPL2 (germ cell), and ALPI (intestinal) (2). Alkaline phosphatase placental-like 2 (ALPPL2) shares high (98%) homology with alkaline phosphatase placental (ALPP), but is uniquely expressed in the thymus, testis, and certain germ cell cancers such as seminoma (3-6). ALPPL2 has become an attractive immunotherapy target due to its restricted expression in healthy tissue and overexpression in various cancers (7). In addition to seminoma, elevated ALPPL2 expression has been identified in gastric and pancreatic cancers and mesothelioma (7-9).
