Species Reactivity: Human Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human OGG1 protein. Background 8-oxoguanine DNA glycosylase (OGG1) is a DNA repair enzyme that repairs oxidative DNA damage. DNA bases like guanine can be oxidized to produce 7,8-dihydro-8-oxoguanine (8-oxoG), the most predominant form of oxidative DNA damage and a biomarker of oxidative stress. If left unrepaired, 8-oxoG can cause mutations during DNA replication. Through its DNA glycosylase and alpha-purine-DNA-glycosylase domains, OGG1 specifically removes 8-oxoG and its open-ring product 2,6-diamino-4-hydroxy-5-formamidopyrimidine from damaged double-stranded DNA (1). DNA damage is implicated in many human diseases, including inflammatory disorders, cancer, and neurodegenerative diseases such as Alzheimer's disease (AD). OGG1 has recently been identified as a potential therapeutic target for AD by promoting DNA repair of oxidative DNA damage that accumulates in aging brain and AD mouse models (2,3). In addition to its role in DNA repair, OGG1 is implicated in various cellular regulatory functions, including transcriptional regulation, inflammation, cell death, and autophagy (4).
