Product Usage Information For optimal ChIP results, use 5 μl of antibody and 10 μg of chromatin (approximately 4 x 10 6 cells) per IP. This antibody has been validated using SimpleChIP ® Enzymatic Chromatin IP Kits. The CUT&RUN dilution was determined using CUT&RUN Assay Kit # 86652 . The CUT&Tag dilution was determined using CUT&Tag Assay Kit # 77552 . Chromatin IP: 1:100 CUT&RUN: 1:100 CUT&Tag: 1:100 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. Background Transcription factor 4 (TCF4), also known as immunoglobulin transcription factor 2 (ITF2), is a member of the basic helix-loop-helix family and interacts with Ephrussi-box (E-box) binding sites (1,2). TCF4 is broadly expressed, and due to alternative splicing, transcripts can give rise to over a dozen different isoforms (3). It plays a key role in neuronal development and has been implicated in a variety of developmental disorders, including Pitt-Hopkins syndrome, schizophrenia, and autism (4-9). In addition to its functions in the brain, TCF4 is essential for the development of plasmacytoid dendritic cells (10-12). Finally, in the context of cancer, TCF4 expression may be associated with divergent patient outcomes; higher TCF4 expression is associated with increased survival in non-small cell lung cancer, yet the protein has also recently been reported to confer immunotherapy resistance in melanoma (13,14).
