Background
A-kinase anchoring protein 12 (AKAP12), also known as Gravin, is a scaffold protein that regulates intracellular signaling by sequestering signaling molecules in subcellular compartments as protein complexes. AKAP12 regulates protein kinase A (PKA) and protein kinase C (PKC)-dependent signaling and controls cell cycle progression, cytoskeletal dynamics, vascular development, glial function, and angiogenesis. AKAP12 expression is correlated with chronic liver diseases (CLDs), inflammatory diseases, and several human cancers (1,2). AKAP12 is a potential therapeutic target for the treatment of esophageal squamous cell carcinoma (ESCC) and colorectal cancer (3,4), and may serve as a biomarker for anti-VEGF therapy-resistant cancer, where its expression is upregulated (5). AKAP12 is highly expressed in heart, regulates desensitization and recycling of β2-adrenergic-receptor (β2AR), and has been implicated in cardiovascular disease initiation and progression (6). Increased expression of AKAP12 in heart is associated with accelerated cardiac dysfunction (7).
