Background Transcription factor A of the mitochondria (TFAM) and transcription factor B2 of the mitochondria (TFB2M) are critical components of the mitochondrial RNA polymerase machinery (1,2). TFB2M binds to POLRMT and induces structural changes, enabling promoter opening (3,4). The ability of TFB2M to bind at mtDNA promoters is regulated by phosphorylation (5). TFB2M has also been shown to have methyltransferase activity towards rRNA (6). TFB2M can be overexpressed in hepatocellular carcinoma, where it promotes growth and metastasis through ROS-Akt-NF-κB signaling (7).
