Product Usage Information Western Blotting: 1:1000 Immunohistochemistry (Paraffin): 1:100 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. Background Epidermal growth factor (EGF) module-containing mucin-like receptor 2 (EMR2), also known as CD312 or adhesion G protein-coupled receptor E2 (ADGRE2), is a member of the adhesion G protein-coupled receptor (ADGRE) family (also referred to as the EGF-TM7 family). This family is a unique class of receptors characterized by a large N-terminal extracellular region containing multiple tandem EGF-like domains, a mucin-like stalk, and a seven-transmembrane domain typical of GPCRs. Similar to other family members, EMR2 plays a functional role in cellular adhesion, migration, and signaling (1). EMR2 is closely related to CD97, also known as ADGRE5, and is likely the product of a gene duplication event; however, EMR2 cannot bind the CD55 ligand that readily binds CD97 (2). EMR2 expression is also restricted to myeloid cells, whereas CD97 expression is relatively ubiquitous (2). EMR2 has been shown to promote differentiation and cytokine release in human macrophages, as well as adhesion, chemotaxis, and degranulation in neutrophils (3-5). EMR2 also contributes to pediatric acute lymphoblastic leukemia (ALL) by promoting an immunosuppressive environment, and its upregulation is associated with poor survival in glioblastoma (GBM) and acute myeloid leukemia (AML) (6,7).
