Background Carnitine palmitoyltransferase-2 (CPT2), localized to the mitochondrial inner membrane, mediates the transport of long-chain fatty acids into mitochondria for β-oxidation (1). Studies show that high-level saturated fatty acid palmitate causes lipotoxicity in liver cells. However, low-level palmitate enhances mitochondrial metabolism and reduces the lipotoxic effect induced by high-level palmitate. The active form of CDK1 is upregulated in the mitochondria of cells treated by low-level palmitate, leading to phosphorylation and activation of SIRT3. The activated SIRT3, in turn, deacetylates and dimerizes CPT2, boosting fatty acid oxidation (2). In addition, transcription factors E2F1 and E2F2 suppress CPT2 expression, generating a lipid-rich environment essential for hepatocarcinogenesis (3).
