Background Haspin (Haploid germ cell-specific nuclear protein kinase, GSG2) is an atypical serine/threonine kinase missing the canonical DFG motif that is present in conventional protein kinases (1). Haspin phosphorylates histone H3 at Thr3 (2). This phosphorylation is important for cell cycle progression, positioning Aurora B kinase at centromeres to regulate centromeric sister chromatid cohesion during prophase and prometaphase, and subsequently ensuring proper chromosome alignment during metaphase and normal chromosome segregation during anaphase (3,4). Haspin has been shown to be overexpressed in multiple human tumor types, with roles in tumor progression and proliferation (5). Inhibition of Haspin kinase is a potential therapeutic approach for cancer treatment (6,7). In addition to cell cycle control, the phosphorylation of histone H3 at Thr3 by Haspin regulates primary cilia length and intracellular homeostasis (8).
