Background Huntingtin's disease (HD) is a neurodegenerative disease pathologically characterized by striatal degeneration and loss of efferent medium spiny neurons. HD is caused by mutations in the gene that lead to additional uninterrupted CAG repeats on the polyglutamine tract of huntingtin protein (1). Huntingtin-associated protein 40 (HAP40), encoded by the gene, is a known binding partner of huntingtin (2). HAP40 acts as a central regulator of huntingtin, necessary for the protein's stability and normal function and has been shown to modulate huntingtin aggregation and toxicity in mouse and models of HD, showing potential for HAP40 as a therapeutic target (3,4).
