Product Usage Information Western Blotting: 1:1000 Immunofluorescence (Immunocytochemistry): 1:1000 - 1:2000 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. This antibody detects both full-length (150 kDa) and processed (105 kDa) LAMC2 proteins. Species Reactivity: Human Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding His747 of human LAMC2 protein. Background Laminins are a family of proteins important for maintaining cellular basement membrane (BM) structure and function (1). Laminins exist as heterotrimers of α (LAMA), β (LAMB), and γ (LAMC) subunits. LAMC2 is the γ subunit of laminin-5 (LN-332 or LN5), which is a triplex composed of laminin α3, β3, and γ2 chains. LN-332 is one of the main isoforms of laminin associated with malignant transformation, proliferation, adhesion, migration, invasion, and metastasis (2). Upregulation of LAMC2 is a prognostic marker for the progression of various cancer types, including colorectal carcinoma, urinary bladder carcinoma, pancreatic ductal adenocarcinoma, and lung carcinoma (3-6). Under physiological conditions, LN-332 locates at the dermal-epidermal junction, provides skin integrity and resistance to external mechanical forces, and promotes adhesion and migration of epithelial cells (7,8). Mutation or deletion of the gene causes Herlitz junctional epidermolysis bullosa, an inherited disease causing severe skin blistering (9,10). The biological activity of LN-332 can be regulated by LAMC2 proteolytic processing through MT1-MMP (MMP14) (11). LN-332 containing full-length LAMC2 (150 kDa) functions to promote adhesion, while LN-332 containing processed LAMC2 (105 kDa) promotes migration (12).
