Background Synaptoporin, also known as synaptophysin II, has been identified as an isoform of synaptophysin in small synaptic vesicles; it shares 58% homology with synaptophysin (1,2). Synaptoporin and synaptophysin are N-glycosylated membrane proteins that are part of a heteromultimeric complex in synaptic vesicle membranes (2). In general, synaptophysin is more widely distributed in most areas of the nervous system, whereas synaptoporin expression is restricted in some subareas, e.g., forebrain and spinal cord (2-4). After peripheral nerve injury, synaptoporin expression is upregulated in small dorsal root ganglion neurons and appears to play a role in both physiological and neuropathic pain states (5).
