Background Transmembrane protein 175 (TMEM175) is a potassium channel located in the membrane of late endosomes and lysosomes. TMEM175 has been shown to regulate lysosomal membrane potential, pH stability, and organelle fusion. In contrast, TMEM175 deficiency impairs lysosomal pH stability, lysosome-mediated autophagosomal clearance, and mitochondrial clearance in cells (1). Dysfunctional mitophagy resulting from TMEM175 loss-of-function mutations has been implicated in several human central nervous system (CNS) diseases, including Alzheimer's disease (AD) and Parkinson's disease (PD) (2-4). Recently, it has been discovered that TMEM175 plays a role in apoptosis, as it is directly inhibited through binding of the anti-apoptotic regulator Bcl-2 (5).
