Background
Ubiquitinating enzymes (UBEs) catalyze protein ubiquitination, a reversible process countered by deubiquitinating enzyme (DUB) action. Five DUB subfamilies are recognized, including the USP, UCH, OTU, MJD, and JAMM enzymes (1,2). Ubiquitin-specific-processing protease 30 (USP30), also known as ubiquitin carboxyl-terminal hydrolase 30, belongs to the USP subclass of DUBs. USP30 is located on the outer mitochondrial membrane, where it regulates mitochondrial dynamics and mitophagy by deubiquitinating outer mitochondrial membrane proteins, such as Tom20 and Miro1. Mitochondrial dysfunction is strongly implicated in the etiology of neurodegenerative diseases, including Parkinson's disease (PD). Knockdown of USP30 increases parkin-mediated mitophagy and clearance of dysfunctional mitochondria, making this protein a therapeutic target for PD. USP30 inhibition was shown to be protective in PD patient-derived neurons and in mouse models of PD, increasing mitophagy and decreasing oxidative stress, phosphorylated alpha-synuclein, and dopaminergic neuron loss. In addition to PD, USP30 is a therapeutic target for various cancers. Its expression is upregulated in many malignancies, while its inhibition has been shown to reduce cancer cell survival and proliferation (3-5).
