Background Cell adhesion molecule 4 (CADM4), also known as immunoglobulin superfamily member 4C (IgSF4C), is an oligodendrocyte-enriched membrane protein. CADM4/IgSF4C is also a member of the cell adhesion molecules (CAMs) family. CAMs are cell membrane glycoproteins that function in cell-cell or cell-matrix interactions. The biological function of CAMs is broad and diverse (1). In the brain, CAMs play a critical role in establishing and maintaining proper neuronal structures and function by mediating extracellular interactions between cells (2). CADM4/IgSF4C is a member of a small group of CAMs (CADM1-CADM4) (3). CADM1-CADM4, also known as synaptic adhesion molecules (SynCam) and nectin-like adhesion molecules (Necl), are characterized by three immunoglobulin-like domains contained in their extracellular region. CADM1-CADM4 likely play an important role in proper myelination of neuronal axons, which is required for proper interneuronal communication, by mediating contact between axons and oligodendrocytes. Altering CADM4 function in oligodendrocytes results in hypomyelination and myelin abnormalities, consistent with the proposed function of these proteins in establishing and maintaining axonal myelination (4).
