Background Synaptojanin (SYNJ) is a phosphatidylinositol (PI) phosphatase located on the plasma membrane (1). SYNJ1 is a brain-enriched presynaptic phosphatase involved in synaptic vesicle recycling, clathrin-coated vesicle uncoating at synapse. While SYNJ1 is brain-specific, its paralog SYNJ2 is ubiquitously expressed. SYNJ2 is also abundantly expressed in the synapse and inhibits clathrin-mediated endocytosis (2). The main function of both SYNJ1 and SYNJ2 is to remove the five-position phosphates from PI (3,4,5)-trisphosphate (PIP ) and PI 4,5-bisphosphate (PIP ) (3).
