Background Ubiquitin-specific-processing protease 20 (USP20), also known as ubiquitin carboxyl-terminal hydrolase 20, is a deubiquitinating enzyme (DUB) and belongs to the USP subclass of DUB (1). Studies show that, after feeding, increased circulating insulin and glucose activate mTORC1, which phosphorylates USP20 at Ser132 and Ser134. The phosphorylated USP20, in turn, is recruited to the HMG-CoA reductase (HMGCR) complex. HMGCR is the rate-limiting enzyme in the cholesterol biosynthesis pathway. HMGCR stabilization through deubiquitination by USP20 in the feeding state, therefore, leads to elevated cholesterol biosynthesis in the liver (2). Furthermore, USP20 was identified as the DUB stabilizing the labile transcription factor SNAI2/SLUG to promote breast cancer metastasis (3).
