Product Description
Monoclonal Antibody for studying KEAP1. Validated for Peptide ELISA (DELFIA). Highly specific and rigorously validated in-house, KEAP1 (D6B12) Rabbit Monoclonal Antibody (Biotinylated) (CST #60857) is ready to ship. Product Usage Information Biotinylated antibodies are ideal for immunoassay technologies and high-throughput ELISA platforms that require antibody pairs where both antibodies are from the same host. Platforms utilizing biotinylated antibodies include, but are not limited to, MSD, xMAP, Quanterix Simoa, AlphaLISA, AlphaScreen, HTRF, LANCE, and TR-FRET. Optimal dilutions/working concentrations should be determined by the end user. Please contact us if you require the antibody clone biotinylated at a different concentration, a carrier-free formulation, or a more customized packaging solution. Storage Supplied in 140 mM NaCl, 3 mM KCl, 10 mM sodium phosphate (pH 7.4) dibasic, 2 mM potassium phosphate monobasic, 2 mg/mL BSA, and 50% glycerol. Store at -20°C. Do not aliquot the antibody. Specificity / Sensitivity KEAP1 (D6B12) Rabbit Monoclonal Antibody (Biotinylated) recognizes endogenous levels of total KEAP1 protein. Species Reactivity: Human, Mouse, Rat Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Glu593 of human KEAP1 protein. Background The nuclear factor-like 2 (NRF2) transcriptional activator binds antioxidant response elements (ARE) of target gene promoter regions to regulate expression of oxidative stress response genes. Under basal conditions, the NRF2 inhibitor INrf2 (also called KEAP1) binds and retains NRF2 in the cytoplasm where it can be targeted for ubiquitin-mediated degradation (1). Small amounts of constitutive nuclear NRF2 maintain cellular homeostasis through regulation of basal expression of antioxidant response genes. Following oxidative or electrophilic stress, KEAP1 releases NRF2, thereby allowing the activator to translocate to the nucleus and bind to ARE-containing genes (2). The coordinated action of NRF2 and other transcription factors mediates the response to oxidative stress (3). Altered expression of NRF2 is associated with chronic obstructive pulmonary disease (COPD) (4). NRF2 activity in lung cancer cell lines directly correlates with cell proliferation rates, and inhibition of NRF2 expression by siRNA enhances anti-cancer drug-induced apoptosis (5). Alternate Names Cytosolic inhibitor of Nrf2; INRF2; KEAP1; KEAP1 delta C; kelch like ECH associated protein 1; Kelch-like ECH-associated protein 1; kelch-like family member 19; Kelch-like protein 19; KIAA0132; KLHL19; MGC10630; MGC1114; MGC20887; MGC4407; MGC9454 Specification REACTIVITY: H M R SENSITIVITY: Endogenous MW (kDa): 60-64 Source/Isotype: Rabbit IgG
