Background Transmembrane protein 106B (TMEM106B) is a type II transmembrane protein with unknown function (1). was first identified as a major risk factor in genome-wide association studies in patients with frontotemporal lobar degeneration (FTLD), a neurodegenerative disorder characterized by atrophy in the frontal and temporal lobes of the brain (2). Localization of TMEM106B in late endosomal/lysosomal compartments suggests a role in regulating endo-lysosomal morphology and function (3). Endo-lysosomal dysfunction is likely a major cellular pathway that contributes to FTLD as other genetically linked FTLD genes, including , , and , converge on this cellular pathway (4). TMEM106B is sequentially processed through ectodomain shedding, followed by intramembrane proteolysis, with possible variability in the intramembrane cleavage site.
