Immunoprecipitation: 1:50 - 1:200
Storage
Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody.
Protocol
Available protocols: Western Blotting, Immunoprecipitation
Specificity / Sensitivity
Phospho-TMEM106B (Tyr50) (F3L6S) Rabbit Monoclonal Antibody recognizes endogenous levels of TMEM106B protein only when phosphorylated at Tyr50. This antibody detects a 110 kDa protein of unknown identity in some cell lines and a 50 kDa protein in A-431 cells.
Species Reactivity: Human, Mouse, Rat
Source / Purification
Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Tyr50 of human TMEM106B protein.
Background
Transmembrane protein 106B (TMEM106B) is a type II transmembrane protein with unknown function (1). was first identified as a major risk factor in genome-wide association studies in patients with frontotemporal lobar degeneration (FTLD), a neurodegenerative disorder characterized by atrophy in the frontal and temporal lobes of the brain (2). Localization of TMEM106B in late endosomal/lysosomal compartments suggests a role in regulating endo-lysosomal morphology and function (3). Endo-lysosomal dysfunction is likely a major cellular pathway that contributes to FTLD as other genetically linked FTLD genes, including , , and , converge on this cellular pathway (4). TMEM106B is sequentially processed through ectodomain shedding, followed by intramembrane proteolysis, with possible variability in the intramembrane cleavage site. Fragments consisting of residues 120-254 of TMEM106B have been shown to form amyloid filaments in human brains (5-7).Â
