Background Prolyl 4-hydroxylase subunit alpha-1 (P4HA1) is a member of the prolyl 4-hydroxylase enzyme family, which is responsible for catalyzing the hydroxylation of specific proline residues within collagen polypeptide chains. This hydroxylation is a crucial step in collagen synthesis and maturation. The hydroxylated proline residues are essential for the stabilization of collagen's triple helical structure by facilitating the formation of hydrogen bonds. This structural stability is fundamental to collagen's mechanical properties and its ability to function as a structural scaffold in various tissues (1). The hydroxylation of proline residues in collagen is also critical for subsequent crosslinking reactions, which further strengthen the collagen fibrils and contribute to the overall stability of tissues. These crosslinks are important for tissue integrity, wound healing, and overall physiological function (1). P4HA1 has recently been identified in gene signatures associated with several tumors (2,3).
