Background Synaptojanin-2 binding protein (SYNJ2BP), also known as OMP25, is a mitochondrial outer membrane protein that acts as a signaling hub, regulating crucial processes in the brain, including neuron function and inter-organelle communication. A key function of SYNJ2BP is managing the contact between mitochondria and the endoplasmic reticulum (ER), known as mitochondrial-ER contact sites (MERCs) (1). Specifically in neurons, SYNJ2BP and Synaptojanin-2 (SYNJ2) form a tethering complex that latches PTEN induced putative kinase 1 (PINK1) mRNA onto the mitochondria. This specialized mechanism enables the local translation of PINK1 in distal parts of the cell, ensuring that distant mitochondria receive a steady supply of the protein required to activate mitophagy (2). By regulating the contact points between the ER and mitochondria in the liver, SYNJ2BP also plays a role in regulating hepatic lipid flux (3).
