Background Piezo1 is a multi-pass transmembrane protein that serves as a mechanosensitive cationic ion channel, directly activated by membrane stretch (1,2). Piezo1 protein is a trimer with three blade-like propellers inserted within the lipid bilayer with a central ion channel that senses various mechanical forces such as membrane tension, shear stress, and cyclical pressure (3,4). Membrane stress induces Piezo1 conformational change, leading to ion pore opening and Ca influx, which further activates multiple related signaling pathways such as CaMKII, NFκB, and Akt (5,6). The protein is engaged in a variety of essential biological processes, including vascular and lymphatic valve development (7,8), musculoskeletal function (9), and bone formation (10). Piezo1 protein plays a regulatory role in macrophage-mediated inflammatory diseases, atherosclerosis, and cancer metastasis (11-14).
