Background Sigma intracellular receptor 2 (SIGMAR2 or TMEM97) is an endoplasmic reticulum (ER)-resident membrane chaperone protein that regulates the sterol transporter NPC1 (1). Recently, SIGMAR2/TMEM97 was identified as the opioid 2 receptor. Opioid receptors are a large super family of 7 transmembrane-spanning G protein-coupled receptors (GPCRs) that are activated by both endogenous opioid peptides and by exogenously administered opiate compounds (2). Opioid receptors are of pharmacological interest not only as analgesics but to treat neurological disorders including psychiatric and neurodegenerative diseases.
