Antibodies are purified by protein A and peptide affinity chromatography. Background N6-methyladenosine (m6A) is a post-transcriptional modification found in various RNA subtypes. Zinc finger CCHC domain-containing protein 4 (ZCCHC4) is an rRNA m6A methyltransferase that specifically carries out m6A methylation of 28S rRNA at position 4220 (1). ZCCHC4 is highly conserved among multicellular organisms, and knockout of ZCCHC4 ablates m6A methylation exclusively at position 4420 on 28s rRNA, suggesting that this is an evolutionarily conserved function (1,2). Knockout of ZCCHC4, and consequently the m6A 4220 modification, has been reported to reduce overall protein translation and inhibit cell proliferation, while others have reported that this knockout had no effect on cell proliferation nor rRNA production (1,2). ZCCHC4 has been found to be overexpressed in hepatocellular carcinoma, and ZCCHC4 knockout cells showed reduced tumor growth in a xenograft mouse model (1). Interestingly, X-ray crystallography reveals that ZCCHC4 contains an autoinhibitory regulatory loop that blocks its catalytic site, a unique feature amongst m6A RNA methyltransferases currently identified (3).
