Background MESH1 (Metazoan SpoT homolog 1), also known as HDDC3 (HD domain-containing protein 3), is an NADPH phosphatase in the cytosol. MESH1/HDDC3 is induced in stressed cells, depleting NADPH and stimulating ferroptosis (1). In addition, removal of MESH1/HDDC3 maintains NADPH levels in cells under stress conditions and therefore promotes ferroptotic cell survival (1). Furthermore, knockdown of MESH1/HDDC3 was shown to induce endoplasmic reticulum (ER) stress and integrative stress responses to facilitate cancer cell survival (2).
