Background Mitochondrial ornithine aminotransferase (OAT) catalyzes the reversible conversion of L-ornithine and 2-oxoglutarate to L-glutamate 5-semialdehyde and L-glutamate (1). Studies show that pancreatic ductal adenocarcinoma (PDA) relies on ornithine synthesis from glutamine, a process mediated by OAT. Ornithine derived from glutamine is used for polyamine synthesis and is critical for PDA growth. In contrast, most adult normal tissues are dependent on ornithine derived from arginine for polyamine synthesis (2).
