Background WD repeat domain 4 (WDR4) protein forms a complex with METTL1 to catalyze 7-methylguanosine (m7G) on mRNA, tRNA, and miRNAs (1,2). METTL1 and WDR4 are required for m7G modification, with METTL1 being the catalytic component and WDR4 stabilizing the complex (2). m7G on mRNA is required for translation and stem cell differentiation, while m7G on miRNA can regulate structure and biogenesis (3,4). mutations impair the methylation of m7G46 of tRNA species and can cause primordial dwarfism and neurological symptoms (5-7). Overexpression of WDR4 has been shown in hepatocellular carcinoma, resulting in increased m7G levels and inhibition of apoptosis (8).
