Background
TRPS1 is a transcription factor that was initially discovered as being mutated in Tricho-rhino-phalangeal syndrome, a craniofacial and skeletal malformation syndrome (1,2). It was also found to be expressed at higher levels in androgen-dependent prostate cancers, reducing the expression of PSA (3,4). TRPS1 recognizes GATA sequences and represses the activation potential of other GATA factors (5). By repressing STAT3 expression, TRPS1 regulates chondrocyte proliferation and survival (6,7). TRPS1 also can increase the activity of HDAC1 and HDAC4 during mitosis to promote histone deacetylation (8). In addition, TRPS1 binds to the NuRD complex via CHD4 and can repress other key transcription factors such as p63 and estrogen receptor (9-11).
