Background Histones are essential proteins that compact eukaryotic DNA into chromatin and are the basis for the epigenetic control of genomic functions. Among the four core histones, the H2A family has the greatest sequence divergence that results in the largest number of variants, such as H2A.X, H2A.Z, and H2A.B, which play various biological functions (1). H2A.B shares only 48% sequence identity with the canonical H2A and is often associated with active gene transcription (2). H2A.B falls into the "short" variant category as it lacks the H2A carboxy terminus, which may notably alter its interactions and dynamic properties within the nucleosome core particle. The H2A.B histone variant appeared in eutherian mammals and is testis-specific (3,4), but it has also been reported to be abnormally expressed in Hodgkin lymphoma cell lines (5,6). Recently, it was demonstrated that H2A.B enhances ribosome biogenesis by increasing transcription of ribosomal DNA and ribosomal protein genes (7).
