Background dUTP pyrophosphatase (dUTPase), also known as deoxyuridine triphosphatase (DUT), catalyzes the conversion of dUTP to dUMP and pyrophosphate (1,2). Studies show that fumarate, an intermediate in the TCA/Krebs cycle, functions as a signaling metabolite to regulate mitochondrial biogenesis. Fumarate binds to the mitochondrial enzyme, malic enzyme 2 (ME2), and stimulates the formation of ME2 dimers. ME2 dimers, in turn, activate the enzymatic activity of dUTPase/DUT, promoting thymidine biosynthesis and increasing mitochondrial DNA replication (3).
