Background Transmembrane protein 115 (TMEM115) is a 35 kDa protein localized to the Golgi cisternae. Both the N- and C-terminal ends of TMEM115 are oriented toward the cytosol, with four transmembrane domains and a Golgi targeting sequence located in between. While the functions of this protein are not fully understood, it is suggested to play roles in Golgi-to-ER retrograde transport and glycosylation (1). Elevated TMEM115 expression levels have been observed in glioma and hepatocellular carcinoma, indicating its potential as a biomarker and therapeutic target for these cancers (2,3). Golgi dysfunction is associated with many diseases, including cancer, neurodegeneration, and cardiovascular diseases. As a Golgi-specific transmembrane protein, TMEM115 can be leveraged for immunoprecipitation experiments to purify and study the Golgi proteome, metabolome, and lipidome in health and disease (4).
