Background
Butyrophilin-like (BTNL) proteins are part of the immunoglobulin superfamily and are structurally related to B7 proteins that comprise co-stimulatory and co-inhibitory molecules that regulate T cells. Five BTNL family members have been identified in humans, and eight in mice (1). BTNL proteins play an important role in regulating γδT cells, and BTNL-fc fusion proteins and soluble BTNL molecules have been shown to inhibit T cell activation (2,3). The BTNL family member, BTNL9, has been identified as a prognostic marker and possible therapeutic target in multiple malignancies, including uveal melanoma, breast cancer, lung adenocarcinoma (LUAD), and thyroid cancer (4-7).
