Background
LPCAT3 (lysophosphatidylcholine acyltransferase 3), also called MBOAT5, is part of the LPCAT family that plays an important role in lipid metabolism (1,2). Its function is to catalyze the incorporation of fatty acids into phospholipids, lysophosphatidylcholine (lysoPC), lysophosphatidylserine (lysoPS), and lysophosphatidylethanolamine (lysoPE), to produce various fatty acid containing forms of PC, PS, and PE. It shows a preference for lysoPC and incorporation of linoleoyl and arachidonoyl chains as fatty acid donors (3,4). LPCAT3 is localized to the endoplasmic reticulum (ER) and is primarily expressed in metabolic tissues, including the liver, pancreas, and adipose tissue (2). It is a direct transcriptional target of LXR, PPARα, and PPARγ, which are lipid activated nuclear receptors that play important roles in regulating lipid homeostasis (2,5-7). Elevated expression of LPCAT3 is associated with the iron-dependent cell death process of ferroptosis that is characterized by an increase in lipid peroxidation (8). LPCAT3 inhibitors have shown a partial protection from ferroptosis (9). Dysregulation of LPCAT3 can have an important role in the development of diseases, including atherosclerosis, diabetes, intestinal tumors, and nonalcoholic steatohepatitis (NASH) (10,11). LPCAT3 is significantly upregulated in many cancers and is associated with prognosis (12).
