This phosphorylation stabilizes the eIF2-GDP-eIF2B complex and inhibits the turnover of eIF2B. Induction of PKR by IFN-γ and TNF-α induces potent phosphorylation of eIF2α at Ser51 (5,6).The eIF2γ subunit is the largest subunit of the eIF2 heterotrimer, and contains the GTP-binding pocket. eIF2γ binds methionylated initiator tRNA in a GTP-dependent manner (7-9). Mutations in eIF2γ cause an X-chromosomal neurological disorder called MEHMO syndrome, resulting in intellectual disability due to impaired translation initiation (10-12).
