Background Microrchidia family CW-type zinc finger 2 (MORC2) is a protein involved in ATP-dependent chromatin remodeling and DNA repair (1-3). Upon DNA damage, MORC2 dimerizes and is phosphorylated by PAK1. PARP1 can then recruit MORC2 to break sites, where it destabilizes histone-DNA interactions, and recruits DNA repair proteins, such as BRCA1 and Rad51 (4,5). MORC2 is recruited by the HUSH epigenetic silencing complex to sites of heterochromatin (6). MORC2 has also been shown to downregulate important tumor suppressors p21 and ArgBP2 in gastric cancers by recruitment of EZH2 (7-9). Mutations in the gene locus have been shown to cause the axonal form of Charcot-Marie-Tooth disease (CMT), a disorder of the motor and sensory neurons of the peripheral nervous system (8-11).
