Background DNA-(apurinic or apyrimidinic site) endonuclease 2 (APEX2/APE2) is an endodeoxyribonuclease that functions in the DNA base excision repair (BER) pathway to repair DNA lesions induced by oxidative and alkylating agents. APEX2 acts in ATR-dependent DNA damage checkpoint activation in response to oxidative stress (1). The gene is synthetically lethal with and genes, which are required for homologous recombination (HR) repair of double-strand DNA breaks. This is due to the ability of APEX2 to reverse single-strand DNA breaks with 3' end blocks. Because BRCA-deficient cancer cells are sensitive to 3' blocks, APEX2 is a potential therapeutic target for HR-deficient cancers (2,3).
