Species Reactivity: Human Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Lys207 of human SPTLC1 protein. Background The serine palmitoyltransferase (SPT) complex catalyzes the initial and rate-limiting step of sphingolipid biosynthesis. This protein complex consists of an SPTLC1/SPTLC2 heterodimer or an SPTLC1/SPTLC3 heterodimer as the catalytic core (1-3). Studies show that mutations leading to unregulated SPT activity and increased sphingolipid synthesis cause amyotrophic lateral sclerosis (ALS), a progressive neurodegenerative disease (4). In addition, mutations in that change the substrate of SPT from serine to alanine drive the accumulation of deoxysphingolipids and cause neuropathy. Furthermore, dietary restriction of serine and glycine also leads to the accumulation of deoxysphingolipids in tumors and reduces tumor growth in mouse xenograft models (5).
