Background
The activating receptor natural killer group 2, member D (NKG2D) is involved in both innate and adaptive immune response, and functions as a switch determining the activation status of natural killer (NK) cells (1,2). NKG2D binds to a diverse family of ligand molecules, which are expressed at low levels in most cells but can be upregulated by cellular stress response (1-3). Retinoic acid early transcript 1 (Raet1) is a GPI-linked cell surface protein that functions as a ligand for NKG2D, and has been shown to activate NK and T cells (3-5). In addition, Raet1 promotes tumor immune surveillance by inducing proliferation of anti-tumor cytotoxic T lymphocytes (3). Raet1 has been shown to be regulated by the STING/cGAS pathway (5), is involved in response to neuronal damage and encephalitis (6,8), and has been implicated in chronic inflammation and autoimmune disease (7,8). The NKG2D-Raet1 interaction is considered a promising target for immune-based therapies due to the specific expression of stress-induced ligands exclusively on tumor cells and strongly induced NK cell activation (9).
