Product Description
Monoclonal Antibody for studying Siglec-10. Validated for Western Blotting,Immunohistochemistry (Paraffin). Highly specific and rigorously validated in-house, Siglec-10 (E1D7S) Rabbit Monoclonal Antibody (BSA and Azide Free) (CST #78255) is ready to ship.
Product Usage Information
This product is the carrier free version of product #74799. All data were generated using the same antibody clone in the standard formulation which contains BSA and glycerol. This formulation is ideal for use with technologies requiring specialized or custom antibody labeling, including fluorophores, metals, lanthanides, and oligonucleotides. It is not recommended for ChIP, ChIP-seq, CUT&RUN or CUT&Tag assays. If you require a carrier free formulation for chromatin profiling, please contact us . Optimal dilutions/concentrations should be determined by the end user. BSA and Azide Free antibodies are quality control tested by size exclusion chromatography (SEC) to determine antibody integrity.
Formulation
Supplied in 1X PBS (10 mM Na 2 HPO 4 , 3 mM KCl, 2 mM KH 2 PO 4 , and 140 mM NaCl (pH 7.8)). BSA and Azide Free. For standard formulation of this product see product # 74799
Storage
Store at -20°C. This product will freeze at -20°C so it is recommended to aliquot into single-use vials to avoid multiple freeze/thaw cycles. A slight precipitate may be present and can be dissolved by gently vortexing. This will not interfere with antibody performance.
Specificity / Sensitivity
Siglec-10 (E1D7S) Rabbit Monoclonal Antibody (BSA and Azide Free) recognizes endogenous levels of total Siglec-10 protein. This antibody also cross-reacts with an unidentified protein of 180 kDa in some cell lines.
Species Reactivity: Human
Source / Purification
Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro673 of human Siglec-10 protein.
Background
Siglec-10 is a sialic acid-binding Ig-like lectin that has been shown to negatively regulate innate and adaptive immune system function (1). Siglec-10 is a type 1 transmembrane protein that has four extracellular Ig-like domains, a transmembrane region, and a cytoplasmic tail with two immunoreceptor tyrosine-based inhibitory motifs (ITIMs) (2). Siglec-10 is expressed in dendritic cells (DCs), monocytes, B cells, NK cells, and T cells (2,3). Siglec-10 interacts with CD24 to suppress immune responses to danger associated molecular patterns (DAMPs) by associating with the tyrosine phosphatase SHP-1, a negative regulator of nuclear factor-kappa B (NF-κB) (4). Siglec-10 has been shown to bind soluble CD52 leading to the impairment of phosphorylation of the T cell receptor-associated kinases Lck and Zap70 and T cell activation (3). It has been proposed that this mechanism of suppression could be involved in T cell homeostasis and the prevention of type I diabetes (3). Siglec-10 has also been identified as the leukocyte ligand for vascular adhesion protein-1 (VAP-1), which plays a key role in leukocyte trafficking (5). Lectin galactoside-binding soluble 3 binding protein (LGALS3BP), a tumor-associated immunomodulatory ligand, has also been shown to bind Siglec-10 (6).
