Background Paired immunoglobulin-like type 2 receptor alpha (PILRA) is an inhibitory surface receptor expressed by myeloid cells and other tissues, including microglia in the central nervous system, regulating immune cells and inflammation (1). It functions by recruiting cytoplasmic phosphatases like PTPN6/SHP-1 and PTPN11/SHP-2, blocking downstream signal transduction via their SH2 domains (2). Furthermore, PILRA plays a role as an entry co-receptor for herpes simplex virus-1 via binding of viral glycoprotein B1 (3). More recently, mutations in the gene have been implicated in Alzheimer's disease (AD) (1,4). Recent large-scale GWAS have linked the risk of developing late-onset AD in humans harboring rare variants of microglial immunoreceptors such as TREM2, CD33, and PILRA (5).
