Background ROD1, also known as PTBP3, is a member of the polypyrimidine tract-binding (PTB) protein family and a critical RNA-binding factor (1). Characterization of the mRNA and protein isoforms of ROD1 has revealed that alternative splicing surrounding the 5′ end of the gene leads to the generation of eight mRNA and three major protein isoforms. Specifically, alternative translation by re-initiation and leaky scanning through downstream start codons leads to the generation of up to three protein isoforms with varying domains and RNA-binding properties (2). ROD1 has been shown to play functional roles in RNA splicing, translational activation, and mRNA stability and is deregulated in numerous cancers, including breast, gastric, and colorectal cancer (3-6).
