Background Diacylglycerol kinases (DGKs) are a unique family of enzymes that phosphorylate the signaling molecule diacylglycerol (DAG) and convert it to phosphatidic acid (PA) (1). DGKs affect various cellular processes by regulating DAG and PA concentrations, including immune signaling, glucose uptake, and neuronal remodeling (2). There are ten DGK isoforms in mammals (α, β, γ, δ, ε, ζ, η, θ, ι, and κ); these isoforms share a catalytic domain but differ in subcellular localization, tissue specificity, and overall function (3). DGK isoform zeta (DGKζ) is one of the primary isoforms expressed in T cells, where it negatively regulates TCR signaling (4). DGKζ attenuates TCR activation by blocking DAG-mediated signaling of downstream effector molecules, which can result in T cell anergy (5,6). Knockout or inhibition of DGKζ has been shown to reverse T cell anergy and enhance the cytotoxicity of chimeric antigen receptor T cells (CAR-Ts), suggesting that DGKζ can be targeted for immunotherapies (7,8). DGKζ may also play a role in cancer, where it is associated with cell proliferation and metastasis in cervical and triple-negative breast cancer (9,10).
