Product Usage Information Western Blotting: 1:1000 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Species Reactivity: Human, Mouse, Rat Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro193 of human MDH1 protein. Background Malate dehydrogenase (MDH) is a key enzyme in the tricarboxylic acid cycle and malate/aspartate shuttle (1,2). MDH is widely expressed in organisms from most bacteria to all eukaryotes (2). The cytoplasmic MDH isoenzyme (cMDH or MDH1) primarily reduces oxaloacetate to malate in the malate/aspartate shuttle (1-3). The major function of the mitochondrial MDH isoenzyme (mMDH or MDH2) is to oxidize malate to oxaloacetate in the tricarboxylic acid cycle (1,2). Studies show that MDH1 is part of the hydride transfer complex (HTC) in the cytoplasm that transfers reducing equivalents from NADH to NADP , reprogramming NAD metabolism and overcoming cellular senescence. HTC thus promotes tumor formation (4). In addition, it was shown that glycolytic activity increases in human breast cancer cells in early hypoxia independent of HIF1α activity. This enhanced glycolytic activity requires NAD provided by lactate dehydrogenase A (LDHA) and MDH1 that is fueled by glutamate-oxoglutarate transaminase 1 (GOT1).
