Background C-type lectin domain family 1 member A (CLEC-1) acts as a pattern recognition receptor for pathogens as a member of the C-type lectin superfamily (1). This protein family plays an important role in innate immunity and the adaptive immune response by stimulating cytokine secretion or T cell priming (1,2). CLEC-1 mRNA is primarily expressed in human endothelial and dendritic cells (DCs) and can be upregulated after stimulation with TGF-β (2,3). CLEC-1 localizes to the endoplasmic reticulum membrane fraction, showing a primarily intracellular localization and function (2,3). CLEC-1 may represent a target for cancer immunotherapy, as it has been shown to act as an immune checkpoint in myeloid cells (4). Absence of CLEC-1 reduced immunosuppressive myeloid cells in tumors and invigorated the activation state of DCs, thereby increasing T cell responses (4).
