Product Usage Information Western Blotting: 1:1000 Flow Cytometry (Live): 1:500 - 1:2000 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. Species Reactivity: Human Source / Purification Monoclonal antibody is produced by immunizing animals with recombinant protein specific to the amino terminus of human CD42b protein. Background The glycoprotein Ib-V-IX (GPIb-V-IX) complex is a platelet receptor that plays a crucial role in hemostasis and general platelet function (1). During vessel injury, multimeric von Willebrand factor (VWF) protein interacts with the GPIb-V-IX receptor expressed on circulating platelets, resulting in platelet activation, aggregation, and blood clot formation (2). GPIb-V-IX is composed of four individual protein subunits: CD42b (GPIbα), CD42c (GPIbβ), CD42d (GPV), and CD42a (GPIX) (3). CD42b, also known as GPIbα and encoded by the gene, is the largest subunit and contains the binding domain for GPIb-V-IX ligands VWF and thrombin (3). Mutations in the gene are often associated with platelet bleeding disorders, such as Bernard-Soulier syndrome (BSS) and platelet-type von Willebrand disease (PT-VWD) (4,5). Autoimmunity against CD42b can result in another bleeding disorder known as primary immune thrombocytopenia (ITP); interestingly, T cells engineered with CD42b chimeric autoantibody receptor (CAAR) have shown promise in specifically targeting autoreactive B cells and treating this disease (6).
