Background NAD kinase 2 (NADK2) is a mitochondrial enzyme that catalyzes the phosphorylation of NAD to NADP . This enzyme uses ATP or inorganic polyphosphate as a phosphoryl donor. Besides NAD , NADK2 can also use NADH as a substrate (1). NADK2 is essential for NADP and NADPH generation in mitochondria. Studies show that tricarboxylic acid cycle activity, mitochondrial folate pathway activity, and oxidative stress in mitochondria are not affected when the gene is deleted. However, deletion leads to diminished NADP and NADPH in mitochondria and negatively impacts the reduction of glutamate to pyrroline-5-carboxylate needed for proline biosynthesis. As a result, deletion slows the proliferation of cells cultured in minimal medium (2).
