Background Polyunsaturated fatty acid lipoxygenase ALOX15, also known as arachidonate 15-lipoxygenase (15-LOX, 15-LOX-1), catalyzes oxygenation of polyunsaturated fatty acids to corresponding hydroperoxides (1,2). Studies show that the small scaffolding protein PEBP1 associates with ALOX15 and polyunsaturated fatty acid lipoxygenase ALOX15B to change their substrate specificity, resulting in the conversion of polyunsaturated phosphatidylethanolamines (PE) to hydroperoxy-PE. Hydroperoxy-PE, when not sufficiently reduced by GPX4, causes ferroptosis (3). In addition, a loss-of-function mutation in ALOX15 leads to protection against nasal polyps and chronic rhinosinusitis (4). Furthermore, ALOX15-positive macrophages contribute to the pathogenesis of a subtype of chronic rhinosinusitis (5).
