Product Usage Information Western Blotting: 1:1000 Immunoprecipitation: 1:50 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Background Transcription elongation factor B polypeptide 2 (TCEB2), also known as Elongin B, is a regulatory component of the SIII/Elongin elongation complex, which regulates elongation by RNA polymerase II by suppressing pausing of the polymerase during gene transcription. The SIII/Elongin complex is also composed of TCEB3/Elongin A, the catalytic subunit, and TCEB1/Elongin C, another regulatory subunit (1). TCEB2/Elongin B is part of the ubiquitin-proteasome pathway as a critical component of multiple E3 ligase complexes. It directly interacts with TCEB1/Elongin C as part of the VHL complex, which targets HIF-1α, a transcription factor that plays a critical role in the cellular response to hypoxia, for degradation (2,3). Dysregulation of HIF-α activity is associated with poor prognosis in multiple human cancers (4). TCEB2/Elongin B promotes therapy resistance in ovarian cancer (5,6), promotes breast cancer progression (7), and reduces radiosensitivity in triple-negative breast cancer (TNBC) (8). During HIV-1 infection, TCEB2/Elongin B is part of an E3 ligase complex that targets the anti-viral proteins APOBEC3F and APOBEC3G for degradation (9).
