Background Non-POU domain-containing octamer-binding protein (NONO), also referred to as p54nrb, is a DNA and RNA-binding protein that is involved in several nuclear processes. NONO binds to the conventional octamer duplex of double-stranded DNA, and can additionally bind to single-stranded DNA and RNA at a location independent of the duplex site (1). NONO belongs to the Drosophila melanogaster behavior, human splicing (DBHS) protein family along with paraspeckle protein 1 (PSPC1) and splicing factor, proline- and glutamine-rich (PSF/SFPQ) (2). NONO, PSPC1, and PSF/SFPQ associate together along with long nonprotein-coding RNA to form ribonucleic protein bodies referred to as paraspeckles, which play a role in gene expression and regulation (2). NONO heterodimerizes with PSF/SFPQ to process pri-miRNA prior to presentation to the Drosha complex (3).
